INVESTIGATION OF THE ANTI-CANCER EFFECTIVENESS OF TELOMERASE INHIBITION WITH BIBR1532 AGENT IN ANAPLASTIC THYROID CANCER CELL LINES
Project no. 22392 · Status: Kapanmış · 2021
Summary
Thyroid cancer (TC) represents one of the most common malignancies of the endocrine system and its incidence has been increasing rapidly worldwide in recent years. Mutations in the telomerase reverse transcriptase (TERT) promoter are among the most prominent prognostic markers of aggression in TCs. The frequency of use of epigenetic drugs is increasing to re-evaluate the possible role of telomere homeostasis in mediating response to cancer treatments and the use of telomerase inhibitors. BIBR1532, a selective anti-cancer agent that inhibits the telomerase enzyme, specifically binds to the active site of hTERT. The hypothesis of our study is that telomerase will be suppressed in thyroid cancer cells and will show a significant anti-cancer activity. In our study, the appropriate number of SW1736 cells was determined for each well with the proliferation assay. The cytotoxic effect of telomerase inhibitor BIBR1532 in SW1736 and C643 thyroid cancer cell lines were evaluated depending on dose and time by WST-1 analysis. Then, the drug was administered at the determined IC50 dose and the cells that did not receive the drug were considered as controls, and apoptosis analysis was performed with Annexin V. The effect of BIBR1532 on the migration rate of cells was investigated by the wound healing test. Finally, the expressions of cancer-related genes were analyzed by real-time qRT-PCR. Treatment with BIBR1532 may be a promising step in the prevention of proliferation and cancer relapse in thyroid cancer correlated with high telomerase activity.
