Project no. 23741 · Status: Kapanmış · 2022
Summary

Saponins; are glycosides with triterpenic, steroidal or alkaloidal aglycones in their structure. In recent years, many different biological activities of them have been reported in scientific literatures. Due to their high complexing efficiency, they also find usages in drug release and drug delivery systems. In this context; in this thesis, studies were carried out on Cephalaria plant species rich in saponins and endemic to our country. For this purpose, firstly, targeted biologically active saponins were obtained from Cephalaria species by different extraction, isolation and purification processes and their structures were elucidated by advanced spectroscopic methods. Advanced spectroscopic techniques such as 1D and 2D NMR and HR-ESIMS were used in structural determination studies. In the second stage of the study, saponin-loaded chitosan nanoparticles (SPCSNP) were synthesized by ionic gelation technique, and the characterization of saponin-loaded chitosan nanoparticles was performed by FTIR, TEM, XPS, SEM and DLS methods. The encapsulation efficiency and particle size of SPCSNP were found to be 38% and 109,04±37,80 nm, respectively. In vitro cytotoxicity studies of SP, CSNPs and SPCSNPs were performed on SH-SY5Y, HL-60, PANC-1, PC-3 and CCD34LU cell lines. In conclusion, saponin-loaded chitosan nanoparticles containing completely natural, biodegradable and biologically active saponins isolated from local and renewable plant resources have the potential to be used as carriers in drug delivery systems.
