Investigation of the Radiosensitizing Effect of the Simultaneous Use of Androgen Receptor Blockers with Ionizing Radiation in Prostate Cancer
Project no. 24012 · Status: Kapanmış (Yayın bekleniyor) · 2022
Summary

Prostate cancer is the most common type of cancer in men worldwide after lung cancer and the second most common cause of cancer-related deaths. The use of radiotherapy with short-term hormone suppression therapy in medium-risk prostate cancer and long-term hormone suppression therapy in high-risk prostate cancer is recommended by all international guidelines. Recently, new studies have come to the fore to intensify hormone suppression treatments. New hormone suppression therapies with proven efficacy are more likely to inhibit androgen receptors. Since it was shown that androgen receptors also send signals for the growth of tumor cells, their simultaneous use with ionizing radiation has become a popular topic. Our aim is to determine cell death and cell cycle changes in prostate cancer cell lines with the simultaneous use of androgen receptor blockers.

Prostate cancer cell lines were used in our study; Before these cell lines were irradiated with ionizing radiation alone, the lethal dose was defined in order to determine the lethal dose of ionizing radiation. The groups created afterwards were ionized radiation + bicalutamide, ionized radiation + enzalutamide, and the last group was ionized radiation + abireterone acetate. Cell lines were compared in terms of cell death parameters and cell cycle changes. The simultaneous use of androgen receptor blockers with ionizing radiation and the differences between each other were evaluated. In our study, it has been shown that the simultaneous use of androgen receptor blockers with ionizing radiation affects cell death, independent of androgen sensitivity. However, in the comparison between these receptor blockers, enzalutamide was found to be the receptor blocker that caused the most cell death with the lowest dose.
