Özet

Rotenone, a commonly used pesticide, has been reported to inhibit the complex-1 enzyme in mitochondria and cause oxidative stress. It is also used to generate toxicity in the cell line. This study was carried out to determine and compare the neuroprotective activity of polyphenols such as curcumin and resveratrol in SH-SY5Y cells that were induced with rotenone. For this purpose, the toxic dose was determined in cell line and the effective dose was determined as 200 nM. 200 nM rotenone was used in the experimental groups. After 24-hour pre-administration of curcumin and resveratrol, rotenone toxicity was established and cell survival was assessed. MTS is a test in which live cells using tetrazolium salts reduce coloration by reducing these salts. MTS cell survival test results show that resveratrol and curcumin have a significant effect on cell survival. As the dose of resveratrol used increased, the protective effect was replaced by toxicity. The clonogenic test results which measures colony forming capacity of cells were evaluated, it was seen that both antioxidants stimulated SH-SY5Y (Neuroblastoma) cell proliferation in all doses. In the final stage of the study, apoptosis was investigated by ELISA method. Although both curcumin and resveratrol decreased the level of active caspase -3, which is an indicator of apoptosis, there was no statistically significant difference in the protection of apoptosis against the control group. Our results showed that rotenone decreased the viability of SH-SY5Y cells in vitro and that antioxidants (resveratrol and curcumin) played a protective role by increasing cell survival and colony forming ability. The caspase-3 mediated apoptosis pathway was not effective in the emergence of these effects. Keywords: rotenone; curcumin; resveratrol; neuronal toxicity; antioxidant
