SYNTHESIS AND CHARACTERIZATION OF DOXORUBICIN LOADED HYALURONIC ACID NANOPARTICLES AND IN VITRO STUDIES OF THEIR APPLICATION FOR DIFFERENT CANCER TYPES
Project no. 633 · Status: Kapanmış · 2017
Summary
According to World Health Organization data, cancer is the second cause of deaths worldwild. Nowadays, lots of different and modern cancer therapy strategies are being developed. Even though current therapy strategies are usually used, they cause too much adverse effects and had restrictions during treatment, thus, new methods for therapy needs to be developed, thus drug delivery systems are also built up. In this study, synthesis and characterization of hyaluronic acid nanoparticles aimed at targeted cancer therapy were carried out and its anticancer properties were provided with loading doxorubicin which is antineoplastic agent. Afterwards, cytotoxicity of synthesized doxorubicin loaded hyaluronic acid nanoparticles was evaluated by performing in vitro studies with different cancer cell lines. Hyaluronic acid nanoparticles couldn’t be prepared with enough yield and quick, thus nanoparticles were synthesized via hyaluronate-chitosan combination. Optimum pH value of prepared chitosan combined hyaluronic acid nanoparticles was found 8. IC50 values of doxorubicin loaded chitosan combined hyaluronic acid nanoparticles were 0.002+/-0.00265 mikro g/ml and 0.00078+/-0.00066 mikro g/ml, and 0.84+/-0.69 mikro g/ml ve 0.29+/- 0.066 mikro g/ml for 48 and 72 hours for SKOV-3 and A549-luc cell lines respectively. As a result, it is considered that doxorubicin loaded hyaluronate nanoparticles would have a potential for being drug carrier for different cancer cell lines.
