Isolation and phylogenetic identification of symbiotic nitrogen-fixing bacteria from Genista spp.

Project no. 75599 · Status: Kapanmış · 2022

Summary

Nitrogen, which is the %78 amount of atmosphere is not a usable form for living organisms. One of the important process of the metabolism for the nitrogen, which is an important element in plant growth, is the symbiotic relationship between Rhizobial species. As a result of this symbiotic relationship, the nodules that growth in plant roots are became a living space for the Rhizobial species and while the nitrogen turn into usable molecular ammonium form in plants. In this study, our aim is phylogenetic identification and isolation the root nodule bacteria from Genista spp. root nodules which is range in the Yamanlar Mountain in Karşıyaka, Izmir province. For this purpose, there is total of 5 root nodules isolated were obtained from G. acanthoclada species to represent different points of Yamanlar Mountain in Izmir. Preliminary grouping of the isolates was using selective-discriminating medium (Congo red YEMA) and then 16S rRNA analysis was performed on the isolates forming light pink-white colonies. Followed by that, analysis of recA, atpD, nodC and nifH gene regions of representative isolates and phylogenetic analysis were done. Regarding to that, this gene regions and the phylogenetic tree's conclusions of them, support each other. Also the isolates of GA7.1 and GA.9 was showed homology with Mesorhizobium genus. Since our study is the first study on root nodule bacteria of G. acanthoclada, the isolates has been introduced to the literature for the first strains representing Rhizobia-Legume symbiotic association.
