Summary

Sea algae are the potential renewable sources of the marine environment, and it is known that these organisms produce various secondary metabolites with large spectrum biological activities. In recent years, the drug active ingredients obtained from marine algae have been interested in. For this reason, many marine permission types are examined in terms of pharmacological properties. Research has been conducted on many marine perpetrators, but after the biological activities have been identified, the wound healing model is very rare. Jania rubens L. is a red sea perception with bioactive compounds, which can be used for various medical and pharmaceutical applications. In this thesis, wound healing activities of J. rubens L. extracts, which have previously been shown to have numerous biological activities but whose wound healing potential has not been established, were evaluated on in vitro and in vivo experimental models. Cell culture studies demonstrated that J. rubens L ethyl acetate and methanol extracts were cytotoxic, and these effects were confirmed by RT-PCR via activation of the Caspase-3 gene. Ethanol and distilled water extracts exhibited higher biocompatibility. Wound closure (scratch assay) experiments showed that certain J. rubens extract concentrations accelerated cell migration and proliferation, thus promoting healing. The in vitro antioxidant activity of some J. rubens extracts provided the first clue that the material may combat oxidative stress at the wound site. In vivo studies were conducted on excisional wound model in rats using a wound dressing contained ethanole extract that demonstrated the most effective results in vitro. Biochemical and histological analyses were conducted on days 7 and 14 following extract application to assess wound closure rates. The results demonstrated that the J. rubens L. extract containing wound dressing modulates the immune response, provides potent antioxidant protection, promotes cell migration and organized tissue synthesis, and orchestrates wound healing through a multifaceted mechanism.
