ISOLATION and STRUCTURALLY IDENTIFICATION of SAPONINS from ENDEMIC YILDIZ PELEMIRI PLANT
Project no. 21173 · Status: Kapanmış · 2019
Summary

In this study, it was aimed to isolate, purify, determine the structure and the biological activity potential of natural saponins from Cephalaria speciosa (Caprifoliaceae) plant. Phytochemical studies on the aerial parts of the plant were resulted with total of 11 saponins 4 of which were revealed as new compounds with 3 new prosapogenins. Total of 6 known saponins namely, macranthoidin A (5), elmalienoside A (6), dipsacoside B (7), decaisoside E (8), scoposide A (9), 3-O-β-D-glucopyranosyl-(1→3)-α-L-rhamnopyranosyl-(1→2)-α-L-arabino-pyranosyl oleanolic acid 28-β-D-glucopyranosyl-(1→6)-β-D-glucopyranosyl ester (10), were isolated. Various chromatographic (CC, VLC, TLC, MPLC) and chemical methods were used during purification and isolation studies. Advanced spectroscopic methods (FTIR, 1D- and 2D-NMR, HR-MS) were used to determine the exact structures of the pure compounds. The cytotoxic activities of the novel saponins and their basic hydrolysis products, prosapogenins were investigated for A549, CCD34-Lu, MDA-MB-231, PC-3, U87MG, HeLa, HepG-2 cell lines. Compound 1, prosapogenins 2 and 4 showed significant cytotoxicity.
