Project no. 22153 · Status: Kapanmış · 2021
Summary

The use of nanostructured materials used for drug delivery and imaging systems in biological applications in the diagnosis and treatment of cancer not only helps the delivery of chemotherapeutic agents to malignant tissues or cells, but also has a very important place in complementary or alternative treatment solutions.

In this project, it is aimed to design a drug-loaded radiolabeled magnetic solid lipid drug delivery system for the diagnosis and treatment of cancer cells. For this purpose, azathioprine (AZA) drug molecule, which is used in the treatment of many diseases with its immunosuppressive effect and is also an anti-cancer agent, was used.

By loading AZA on the synthesized magnetic solid lipid nanoparticles, radiolabeling study with technetium-99m was carried out and the biological behavior of this radiolabeled nanocomposite material was investigated in vitro.

Structural characterization of synthesized magnetic solid lipid nanoparticles (SLMNP) was performed by Dynamic Light Scattering (DLS) method, Zeta Potantial Analysis, Scanning Electron Microscopy (SEM) imaging. Azathioprine loaded magnetic solid lipid nanoparticles were radiolabeled with technetium-99m. Quality control studies of azathioprine loaded radilabeled magnetic solid lipid nanoparticles (99mTc-AZA-SLMNP) were performed by Thin Layer Radiochromatography (TLRC) and dose calibrator methods. Magnetic solid lipid nanoparticles have been synthesized via hot homogenization technique. A trial of azathioprine loading was performed on SLMNP structures with three different drug ratios and highest yield has been found as 86 % with 2,5 mg azathioprine ratio via High Liquid Chromatography (HPLC). After labeling with 99mTc, radioactivity measurement was taken with the dose calibrator. The radiolabeling yield was obtained as 90.3% for AZA-SLMNP.

The biological behavior of azathioprine loaded radiolabeled magnetic solid lipid nanoparticles (99mTc-AZA-SLMNP) was monitored in vitro in HaCaT and U87 cell line.
