EVALUATION OF IN VITRO ANTICANCER EFFICIENCY OF ANTI-MIR SPECIFIC TO MIR-146A, MIR-155, MIR-181A AND PREDNISOLONE APPLICATIONS IN CHILDHOOD ACUTE LYMPHOBLASTIC LEUKEMA CELL LINES
Project no. 64 · Status: Kapanmış · 2016
Summary
Prednisolone has been used frequently in the treatment of acute lymphoblastic leukemia since the mid-1950s. However, in order to overcome the problems such as relapse or drug resistance, the development of additional therapies is of great importance. Short, non-protein-coding RNAs, miRNAs, are critical epigenetic regulators with physiological and pathological importance. The aim of this study was to determine the molecular efficacy of miR-146a, miR-155, miR-181a and let-7e inhibition on both leukemia and healthy cells, individually and in combination with prednisolone. For this purpose, cytotoxicity, proliferation and cell cycle, apoptosis tests were performed by using WST-1, trypan blue, APC-BrdU, Annexin V and JC-1 methods in each application group, respectively. The results showed that anti-miR application was effective on the viability, proliferation, cell cycle and apoptosis of leukemia cells and this effect was increased with prednisolone administration. In addition, this activity was found to be very low in healthy cells. In conclusion, it has been shown that anti-miR applications have the potential for clinical use adjuvant to or as an alternative to conventional approaches.
