Preparation, Development, In Vitro Antimicrobial Activity and Cytotoxicity Evaluation of Doxycycline and Atarovastatin Loaded Carboxymethyl Chitosan Hydrogel for the Treatment of Periodontal Diseases

Project no. 29574 · Status: Kapanmış (Yayın bekleniyor) · 2023

Summary

Periodontal diseases are inflammatory diseases that occur due to the interaction between microbial dental plaque and the host response. The traditional treatment of periodontal diseases is nonsurgical periodontal treatment, which includes tooth surface cleaning and root surface planing. However, even if perfect oral care and ideal periodontal treatment conditions are provided, it is not possible to achieve the desired healing conditions in every case and every region. Localized factors such as root anomalies, very deep pockets and furcation involvement, especially together with systemic factors such as diabetes and smoking history, are the limiting factors of the treatment. In such cases, the use of chemotherapeutic agents has been on the agenda in the clinic and literature for many years. Although the oral use of systemic antibiotics provides improvement in periodontal clinical parameters, it also brings various disadvantages. It is difficult for antimicrobials used systemically to reach the gingival crevicular fluid and be in a concentration where they will be effective. In addition, gastrointestinal system irritation and development of antibiotic resistance can be considered among the disadvantages of systemic antimicrobials. One way to benefit from the positive effects of chemotherapeutic agents while eliminating their negative effects is localized drug delivery systems. Carboxymethyl chitosan has the expected properties such as mucoadhesiveness, non-toxicity, and antimicrobial properties as a drug carrier. It is also thought that the multifaceted pharmacological approach in the treatment of periodontitis, which is a multifactorial disease, will improve the treatment results. In the formulation to be developed, it is aimed to benefit from the antimicrobial, anticollagenolytic effect, suppression of bone destruction, and anti-inflammatory properties of doxycycline, and the anti-inflammatory, immunomodulatory, antioxidant, and bone formation stimulating properties of atorvastatin. The synthesis, characterization, and drug release properties of the formulation to be prepared in the form of hydrogel will be evaluated in vitro. The antimicrobial activity of the developed formulation on Escherichia coli, Staphylococcus aureus, and Porphyromonas gingivalis will be evaluated under in vitro conditions. The antimicrobial activity of the developed formulation on Escherichia coli, Staphylococcus aureus and Porphyromonas gingivalis under in vitro conditions and also its cytotoxicity on human keratinocytes will be evaluated.
