SCREENING OF BACTERIAL LIPASE PRODUCERS AND OPTIMIZATION OF PRODUCTION MEDIA
Project no. 25242 · Status: Kapanmış (Yayın bekleniyor) · 2022
Summary

The project aims to screen bacterial lipase producers, determine the bacterial strains exhibiting the highest lipase enzyme activity, conduct optimization studies for production media, and perform a 3L fermenter study with the selected bacterial strain. This study examined lipase enzyme production in the bacterial strains of Bacillus cereus RSKK 2027, Bacillus megaterium RSKK 278, Bacillus pumilis NCT 8241, Bacillus subtilis RSKK 2012, Pseudomonas aeruginosa ATCC 27853, Pseudomonas fluorescens RSKK 10011, Micrococcus luteus RSKK787, and Staphylococcus aureus ATCC 29213. As a result, Bacillus sp. and Pseudomonas sp. bacterial strains were selected, and subsequent evaluations of lipase enzyme activity led to the selection of Pseudomonas sp. strains for the optimization of medium experiments. Consequently, statistical experimental design methods were used to optimize the production medium for the bacterial strains P. fluorescens RSKK 10011 and P. aeruginosa ATCC 27853. Following the optimization of the production media, due to the higher lipase activity, Pseudomonas sp. was used for the 3-L batch bioreactor production and large-scale parameter variations were analyzed.
