Summary

Cancer is the second most common cause of death worldwide after cardiovascular diseases. Chemotherapy is a widely used treatment method, but serious side effects can limit its effectiveness. Natural compounds, due to their low toxicity and diverse mechanisms of action, hold significant potential as alternative treatment options. Drug delivery systems are becoming increasingly important to enhance the therapeutic efficacy of these compounds. Thymoquinone (TQ) is a natural compound found in Nigella sativa with notable anticancer properties. However, its bioavailability is limited due to its poor solubility in water. In this study, the aim was to enhance the therapeutic efficacy of thymoquinone for use in the treatment of various cancer types and to achieve an effective and safe therapy at low doses. Accordingly, thymoquinone-loaded, alginate-coated PLGA (poly D, L-lactic-co-glycolic acid) nanoparticles were prepared using the nanoprecipitation method. Prepared nanoparticles were analyzed by HPLC method; encapsulation efficiency and drug release profile were investigated. Additionally, their chemical structure, morphology, and particle characteristics were characterized through FTIR, SEM, and TEM analyses.

The findings show that the developed nanoparticles can potentially increase the bioavailability of thymoquinone by providing controlled release and can be a promising drug delivery system in cancer treatment.
