Preparation of Radiolabeled Human Serum Albumin Nanoparticles for Cancer Diagnosis
Project no. 253 · Status: Kapanmış · 2016
Summary
The aim of the present study is to develop drug delivery systems with methotrexate (MTX) loaded human serum albumin (HSA) nanoparticles for cancer diagnosis. Within the scope of our project, ISA nanoparticulate formulations were prepared to be used for breast cancer diagnosis. The prepared formulations were evaluated in the terms of particle size, PdI values and zeta potentials and formulations not suitable for use were excluded from the study. MTX was loaded with different ratios to ideally determined formulations. After, the prepared formulations were evaluated in the terms of particle size, PdI values and zeta potentials and formulations not suitable for use were excluded from the study. Ideal formulations (with or without MTX) were placed in stability cabinets for long-term stability study. Formulations intended for use in the diagnosis of breast cancer were labeled with 99mTc radionuclide using stannous chloride as the reducing agent and the labeling efficiency of the nanoparticles was investigated. As a result of radiolabeling studies, it was determined that all formulations were radiolabeled with high labeling efficiency. In vitro cell culture studies were carried out with ISA nanoparticles with and without MTX, to shed light on the in vivo behavior of newly developed formulations and cell incorporation ratios of formulations were investigated on MCF-7 (breast cancer) and HaCaT (human keratinocyte) cell lines. At the end of the studies, it was determined that each of the formulations with and without MTX have 2 times less than target/non-target uptake ratio but have a significantly higher uptake than R/H 99mTc.
