DEVELOPMENT OF INNOVATIVE BIO ANALYSIS SYSTEMS FOR DOPING USED ANALYSIS AND VERIFICATION BY LC-QTOF / MS

Project no. 1114 · Status: Kapanmış · 2018

Summary

Within the scope of the project, it is aimed to design test kits that can enable the on-site analysis of doping agents and to optimize methods to verify by chromatographic methods. In the project, test surfaces were created for innovative biosensor methodologies for DHEA-S and testosterone doping agents selected as models using the appropriate surface chemistry, and their performance was investigated by applying doping standards to the test. As the DHEA-S biosensor system, antibody-based electrochemical biosensor system was developed within the scope of the project. Gold electrodes are used for this system. Electrode surfaces were first coated with cysteamine, then DHEA antibodies were immobilized to the surface using glutaraldehyde as a crosslinker. As for the testosterone detection, the antibody-based electrochemical biosensor system was built on surface-imprinted carbon electrodes (SPE). The SPE surface was coated with magnetic nanoparticles modified with testosterone antibodies using glutaraldehyde as a crosslinker, and capture of testosterone analyte was achieved. Modification steps of both systems were monitored and approved by electrochemical techniques. The linear working range for the DHEA-S biosensor system is 2.5-100 ng / mL and the detection limit (LOD) is 3.971 ng / mL. The linear working range of the testosterone biosensor system is 25-1000 ng / mL, and the LOD value is 27.00 ng / mL. Interfering molecules such as cholesterol, DHEA, bovine serum albumin were used to determine the selectivity of the developed sensor systems. In addition, for both biosensor systems, experiments were carried out with standard addition method in synthetic serum and urine samples and successful results were obtained. Biosensor systems that are practical, innovative, cost-effective and portable designed for the analysis of doping substances are the outcomes of the project.
