Investigation of the Effects of the EZH2 Inhibitor Tazemetostat on Apoptosis and Wnt Signaling Pathways in Anaplastic Thyroid Cancer Cells

Project no. 27364 · Status: Kapanmış (Yayın bekleniyor) · 2023

Summary

Anaplastic thyroid cancer (ATC) is one of the rarest yet most fatal types of thyroid cancer, with a high mortality rate and a life expectancy typically limited to only a few months after diagnosis. In ATC, the EZH2 protein stands out due to its overexpression, which promotes cell proliferation and metastatic processes. In recent years, the EZH2 inhibitor Tazemetostat has shown promise as an anticancer target, especially in cancers with high proliferation rates and resistance to treatment. This study investigates the potential anticancer effects of Tazemetostat and gene expression changes in the Wnt signaling pathway using the SW-1736 cell line as an ATC model.

Following Tazemetostat treatment, significant gene expression changes were observed in the cells; after 24 hours, some genes such as DKK3 and DAAM1 were upregulated, while others like CCND2 and PITX2 were downregulated. By the 48- and 72-hour treatment marks, notable expression changes persisted in certain genes associated with the Wnt signaling pathway. These findings suggest that the EZH2 inhibitor may restrict ATC cell proliferation and potential metastatic activity by repressing or activating specific genes in the Wnt signaling pathway.

The study’s findings indicate that Tazemetostat exhibits pronounced anticancer effects linked to the Wnt signaling pathway and could serve as a novel therapeutic strategy for ATC treatment. With further research, these results suggest that EZH2 and the Wnt signaling pathway may become significant targets in ATC therapy, providing innovative contributions to the clinical management of the disease.
