Summary
EFFECT OF HYDROXYPROPYL-ß-CYCLODEXTRIN (HP-ß-CD) AND HP-ß-CD LOADED POLYMERIC NASAL MICROSPHERES ON THE EXPRESSIONS OF SOME PRO-INFLAMMATORY AND CAV-1 mRNAs IN EXPERIMENTAL MODEL OF ALZHEIMER’S DISEASE INDUCED BY AMYLOID-ß PEPTIDE (1-42) The accumulation of amyloid (Aß) plaques in the brain is one of the key events that play a role in the neuropathogenesis of Alzheimer's disease (AD). One of the factors that cause the development of the disease in this process is neuroinflammation. Targeting "lipid rafts" in neuronal membranes and altering cholesterol levels in these structures are thought to be a neuroprotective approach for AD because of its causal role in the disease mechanism-induced by Aß. It has been suggested that 2-hydroxypropyl-ß-cyclodextrin (HP-ß-CD), a cyclodextrin (CD) derivative, can selectively alter the level of membrane cholesterol. In this study, we aimed to determine the neuroprotective effect of hydroxypropyl-ß-cyclodextrin (HP-ß-CD) and HP-ß-CD loaded polymeric nasal microspheres treatments on the Alzheimer's disease model, which was conducted by stereotaxic administration Aß-peptide (1-42) into the rat brain. In this frame, expressional levels of Cav-1, a cholesterol-binding protein and component of lipid raft structures, and NF-?B mRNA expression levels as a major neuroinflammation marker have been determined in brain cortex tissues using a number of basic molecular biological techniques. According to our results, Aß (1-42) treatment reduced Cav-1 mRNA expression levels and increased NF-?B mRNA expression levels in rat brain cortex. Treatments with HP-ß-CD and HP-ß-CD loaded polymeric nasal microspheres against Aß (1-42) increases Cav-1 mRNA expression level and decreases NF-?B mRNA expression levels. As a conclusion, it is possible to say that HP-ß-CD can be considered as a potential neuroprotective approach to AH. Keywords: Alzheimer’s Disease, 2-hydroxypropyl-ß-cyclodextrin ,Cav-1, NF-?B
