Summary

Objective: Despite advances in cancer diagnosis and treatment, infections remain among the leading causes of morbidity and mortality in cancer patients. Bacteria can contribute to cancer through genetic and epigenetic alterations, potentially accelerating progression and altering treatment response. This study aimed to investigate the effects of bacteria on cancer development and treatment. Methods: An infection model was established by infecting A549 lung cancer cells with Staphylococcus aureus (S. aureus) 6538P. The effects of carboplatin and doxycycline on A549 cells and S. aureus were determined using XTT and MIC assays. The selected drug doses were applied to infected cells, and the expression of cancer-related genes was analyzed via RT-qPCR. In addition, cell cycle, migration, and DNA damage assays were performed to assess cellular responses. Results: The infection model was successfully established, and the presence of intracellular and extracellular S. aureus was confirmed by microscopic staining. Carboplatin was found to be effective at 1000 µM in A549 cells, while doxycycline showed no cytotoxic effect. The MIC value of doxycycline against S. aureus was 0.9765 µg/ml, and the minimum bactericidal concentration was 625 µg/ml. Carboplatin had no inhibitory effect on the bacteria. In infected A549 cells, proliferation and migration significantly increased, while carboplatin's effectiveness was reduced. Cell cycle analysis revealed a decrease in the G1 phase and an increase in the S and G2 phases. No significant DNA damage was observed in response to infection. Conclusion: S. aureus infection enhances proliferation and migration in lung cancer cells and reduces the efficacy of carboplatin. These findings suggest that bacterial infections may play a critical role in cancer progression and treatment response and should be taken into consideration during therapy.
