ASİMETRİK DİMETİLARGİNİN (ADMA) APTAMERLERİNİN GELİŞTİRİLMESİ VE APTAMER TEMELLİ TAYİN KİTİNİN TASARLANMASI

Project no. 20336 · Status: Kapanmış · 2017

Summary

Asymmetric dimethylarginine (ADMA) is known to be associated with many pathophysiological conditions including cardiovascular diseases and therefore studies on this molecule gradually increase. Detection of ADMA as a new approach for diagnosis, treatment and control of these diseases has gained importance. Since ADMA detection methods are limited and insufficient, new methods are needed. High cost of the existing ADMA determination methods, long analysis time and difficulty in using them for routine analysis increase the need for new methods. Nucleic acid aptamers are single-stranded DNA or RNA molecules that provide superiority to antibodies in terms of many properties such as stability, ease of synthesis, and cost. In this respect, aptamers are ideal molecules for design of novel kits as alternative to ELISA. In this project, design of a new method for detection of ADMA, an amino acid derivative with biomarker potential, was aimed. For this purpose, it was aimed to develop ADMA aptamers and then to design of a fluorescence based detection kit. In the context of this project, ADMA aptamer was developed for the first time in the literature by using SELEX method. Then, different fluorescence-based methods were compared for future kit design. Among these methods, the 3 '-FAM labeled aptamer produced a fluorescence response and the required assay for kit was obtained.
