Project no. 29658 · Status: Kapanmış (Yayın bekleniyor) · 2023
Summary

Plasmid isolation is one of the fundamental techniques in molecular biology and life sciences research. The primary objective of current plasmid DNA (pDNA) isolation methods is to obtain plasmids with high purity and yield. However, while manual methods are more cost-effective, they often fail to achieve the desired purity levels in sensitive applications such as cloning and mammalian cell transfection. Consequently, researchers in fields including microbiology, molecular biology, biotechnology, and immunology frequently prefer commercial kits that provide highly pure pDNA. Nevertheless, the majority of these kits are imported, and their high cost poses a significant disadvantage by limiting the efficient use of research budgets in our country. This project proposal aims to eliminate this drawback by developing locally produced mini-, midi-, and maxi-scale pDNA isolation kits for use in life sciences research.

Existing pDNA isolation kits are generally based on silica particles or silica membranes. In contrast, the proposed model will introduce pDNA isolation kits utilizing glass fiber material, applicable in both column and tube formats. The cost of glass fiber is considerably lower compared to commonly used silica-based columns. Moreover, no product based on glass fiber has previously been introduced for pDNA isolation. In laboratory-scale preliminary studies, prototype columns designed with glass fiber were tested, and they successfully provided high-purity and efficient isolation suitable for various applications. These findings suggest that the kits to be developed will offer a cost advantage compared to existing commercial products and reduce dependence on imported kits. In conclusion, the development of plasmid isolation kits with broad applicability across different research fields through local production will not only provide a significant cost advantage but also contribute strategically to strengthening our country’s biotechnology infrastructure.
