Development of Nanomaterial Based Bioactive Dentin Adhesive Systems

Project no. 23876 · Status: Kapanmış (Yayın bekleniyor) · 2022

Summary

Aim: The objective was to assess the cytotoxicity, physical characteristics, and antibacterial attributes of a universal self-etch adhesive containing Arg-greft-p(HEMA) nanoparticles at various concentrations.

Methods: After synthesizing and characterizing the Arg-greft-p(HEMA) nanomaterial, it was incorporated into a universal self-etch adhesive, specifically OptiBond Universal by Kerr Dental, at concentrations of 0%, 5%, 10%, 15%, and 20%. Cytotoxicity of the adhesive, applied to square dentin samples and tested against the L-929 cell line, was determined through cell culture/extraction and MTT assays, with absorbance readings taken using a UV-vis spectrophotometer. Antibacterial properties against S. Mutans and S. Sanguis were assessed through a disc diffusion method, measuring inhibition zones after 48 hours. ATR-FTIR spectroscopy was employed to gauge the degree of conversion in the adhesives, calculated based on aliphatic and aromatic carbon-carbon double bond ratios in the absorbance spectra obtained before and after light polymerization of the adhesives. Statistical analysis was performed using SPSS 25.0 software, with significance set at p<0.05.

Results: Cytotoxicity of the adhesives increased with higher concentrations of Arg-greft-p(HEMA). Antibacterial activity against S. Mutans and S. Sanguis also rose with increasing concentrations of Arggreft-p(HEMA). ATR-FTIR analysis revealed a lower degree of conversion as the concentration of Arggreft-p(HEMA) increased.

Significance: A universal self-etch adhesive containing Arg-greft-p(HEMA) nanomaterial at concentrations of 5% to 10% exhibited strong antibacterial properties while maintaining acceptable physical and cytotoxic characteristics. Further research is needed to support the application of different Arg-greft-p(HEMA) concentrations within the 5% to 10% range.

Keywords: Self-etch dentine bonding agent, universal dentin bonding agent, HEMA, Arginine, nanoparticle, antibacterial adhesive, bioactive dentin bonding agent
