DEVELOPMENT OF ELECTROCHEMICAL IMPEDANCE SPECTROSCOPY BASED SENSOR, WITH HYDROXIDEOXYGUANOSINE IMPRINTED AND INCREASED SELECTIVITY POWER WITH DIDEOXYCYTOSIN

Project no. 21888 · Status: Kapanmış · 2020

Summary

Currently, there are limitations to the determination of 8-hydroxyguanosine (8-OHG), which serves as an important biomarker of oxidative stress. The LC-MS/MS method provides accurate results at femtomolar levels from the genomic DNA obtained in tissue, plasma and urine samples. The most important limitation of this method is the necessity of DNA isolation before analysis, a step which requires the need for expert personnel and extra consumables. It has also been suggested that all analytical methods requiring DNA isolation during the purification process may contribute to total damage by initiating additional DNA oxidation. In the current project, to overcome these limitations, we used a completely innovative method to develop a sensor for the determination of 8-OHG. Considering the chemical structure of the molecule and its potential non-covalent bonding behaviors, we designed aspecial electrode with molecular imprinting technology (MIT). 8-OHG measurement was carried out impedimetrically with high performance parameters (R2 value in the range of 500-10 000 pg/mL, 6 minutes). 0.9928 ± 0.006, LOD 155.8 pg/mL and LOQ 472 pg/mL). In this way, a sensitive, low-cost, fast and innovative technique with higher selectivity has been introduced. We believe this new sensor can provide important potential gains which will help us in the diagnosis and follow-up of oxidative stress-related diseases.
