Development of Analytical Method for The Determination of the Nicotine Metabolite Ratio and Investigation of the Relationship of This Rate with Tobacco Use Levels in Individuals with Tobacco Use Disorders

Project no. 22799 · Status: Kapanmış (Yayın bekleniyor) · 2021

Summary

Nicotine in tobacco is an addictive substance. It is known that nicotine addiction is the primary reason for the continuation of smoking behaviour and the general failure of treatment attempts. Presenting concrete laboratory data specific to individuals in smoking cessation treatments contributes to increasing the success of smoking cessation. In this context, understanding metabolic differences can provide more effective management of individual treatment needs.

This study, it is aimed to lay the groundwork for metabolic differences that play an active role in nicotine metabolism to contribute to the development of a personalized treatment plan with laboratory data that can provide support data for treatment. In the first stage of our project, individuals who applied to Ege University Substance Abuse Toxicology and Pharmaceutical Sciences Institute Smoking Cessation Polyclinic were included. Nicotine Addiction Fagerström Test (FNBT) was applied to the individuals participating in the study. In the second stage, nicotine and its metabolites were extracted by developing the QuEChERS technique to be applied to the urine samples taken from the individuals selected for the study to calculate the nicotine metabolite ratio, which is a phenotypic surrogate of nicotine exposure, to improve the results of smoking cessation. Samples were analyzed by a validated method using gas chromatography-mass spectrometry. The R2 value of the method was >0.99, and linear results were seen in the 25-2000 µg/L working range.

In this study, presenting supportive enzyme activity data to leave a difficult situation such as addiction for the clinician will contribute to the motivation to increase both the success of the treatment and the compliance of the patients with the treatment.
