COMPARATIVE ANALYSIS OF TUMOR CELL, ROOT CELL AND SOMATIC CELL LINES WITH ATOMIC FORCE MICROSCOPE (SPM) AND MOLECULAR BIOPHYSICAL METHODS
Project no. 158 · Status: Kapanmış · 2018
Summary
While embryonic stem cell and cancer cell biology are known to have many similarities in common signalling; healthy somatic cells are known to be different in many ways. Any characterisation of embryonic stem cell is crucial for cancer development and cancer recurrence due to shared signalling pathways and life course with cancer initiator and cancer stem cells. Besides, it is necessary to reveal the relevance between embryonic stem, cancer and somatic cells because the sources of the somatic and cancer cells are mouse embryonic stem cells. With the increasing importance of multidisciplinary studies nowadays, the reflection of the physical and chemical phenomena occurring on the cell biology basis has also gained importance. In this project, we continued our previous molecular biological studies to widen our understanding in physical view with atomic force microscopy, which enables us to take topological measurements from biological samples. We examined mouse embryonic stem cells, mouse lung squamous cancer cells and mouse skin fibroblast cells; Human GBM cells and human astrocytes topologically with atomic force microscope by three different parameters. As a result of this study, roughness topology of the embryonic stem cells and cancer cells were similar and somatic cells were found to statistically different. Hence, these techniques should be considered use in cancer and stem cell characterization and may be candidate diagnostic tool in medicine. The aim of the project is to determine the points where mouse origin cancer cell, somatic cell and stem cell groups are different or similar to each other by Atomic Force Microscopy (SPM) and X-Ray Photoelectron Spectroscopic (XPS) analysis and to reveal therapeutic targets in the case of cancer cells.
