Identification of the association between the development of glaucoma and TGFβ1 -509C>T promoter region polymorphism

Project no. 23307 · Status: Kapanmış (Yayın bekleniyor) · 2022

Summary

Aim: The purpose of this study was to investigate the potential association of the TGFβ1 -509C>T polymorphism with the development of Primary Open-Angle Glaucoma (POAG) and Primary Angle-Closure Glaucoma (PACG) in Turkish population.

Methods: This study included three groups consisting of patients from the Ophthalmology Departments of İzmir Katip Çelebi University and Ege University: a POAG group (n=115), a PACG group (n=53), and a control group (n=96). Detailed eye examinations were performed, and peripheral blood samples were collected for DNA isolation. The TGFβ1 -509C>T polymorphism was evaluated in all participants via PCR amplification using custom-designed primers, followed by sequencing analysis on the Illumina Miniseq platform.

Results: No statistically significant difference was found among the groups regarding demographic characteristics. POAG and PACG groups showed significant differences in intraocular pressure and cup/disc ratio compared to the control group. However, no significant association was found between TGFβ1 -509C>T polymorphism and the development of either POAG or PACG in terms of allele frequency, genotype, and dominant/recessive model analysis.

Conclusion: In a large cohort of Turkish subjects, the TGFβ1 -509C>T polymorphism was not associated with the development of either POAG or PACG. Further comprehensive studies reflecting the broader Turkish community are needed to corroborate these findings.

Keywords: DNA analysis; Glaucoma; Polymorphism; TGFβ1 gene
