Summary
Asymmetric synthesis is one of the most important research areas in organic chemistry and pharmacology. Because chiral compounds have a special place in the pharmaceutical industry. The importance of the relationship between pharmacological action and chirality emerged in the 1960s when thalidomide was administered to pregnant women. While (R)-thalidomide showed anti-nausea properties, the (S)-enantiomer caused birth defects. The importance of chiral compounds is increasing day by day, so studies on the synthesis of these compounds are accelerating. Asymmetric synthesis is the most important method used in the preparation of chiral compounds. In this method, the desired enantiomer of high purity can be obtained by using chiral catalysts. N-heterocyclic carbenes (NHC) are widely used ligands in transition metal compounds. They form more stable complexes against air, moisture and heat compared to NHC phosphine analogs and are used as catalysts in many chemical reactions due to these properties. Therefore, the development of chiral derivatives of these ligands in asymmetric catalysis has also been inevitable. Their low toxicity, easy and economical synthesis are among the reasons for this interest. For this purpose, in this project, chiral sugar-based NHC ligands and their metal complexes were carried out in situ for asymmetric synthesis. The structure and purity of the metal complexes of the sugar-based N-heterocyclic carbene ligands synthesized in the project were confirmed by 1H and 13C NMR, IR, UV, X-Ray, melting point and optical rotation angle analysis methods. These synthesized N-heterocyclic carbene (NHC) complexes were used as catalysts in asymmetric transfer hydrogenation reactions of ketones and their catalytic activities were investigated. Gas Chromatography (GC) was used for the catalytic tests. In addition, the anticancer activities of synthesized sugar-based chiral NHC ligands will be examined using the widely used HeLa human uterine cancer cell line and A549 lung cancer cell line and MTT cell proliferation method in further activity studies.
