DETERMINATION OF THE EFFECTS OF RESVERATROL ON APOPTOSIS IN INTESTINAL CELLS IN THE EXPERIMENTAL NECROTIZING ENTEROCOLITIS CELL CULTURE MODEL
Project no. 53 · Status: Kapanmış · 2016
Summary
Necrotising enterocolitis is a spectrum of ischemia in bowel wall from focal mucosal injury to total ischemia of the whole bowel which is caracterized with a systemic inflammatory response initiated with intestinal mucosal immune system in premature newborn. Because of its high morbidity and mortality, clinical and experimental studies are continuing intensively. The aim of our study was to to evaluate the effects of resveratrol on apoptosis of intestinal epithelial cells in a cell culture model of necrotising enterocolitis. IEC-6 cells were incubated with resveratrol at 3 doses (0,5/1/5 µM). After an hour, inflammation was induced using lipopolysaccharide (LPS) or a cytokine mixture (TNF-? 10 ng/ml; IFN-?: 100 ng/ml and IL-1ß: 1ng/ml). Wounds were constituted on confluent cells and progress of cell migration into the wound was photographed at 0, 2, 4, 6, 8, 10, 12 and 24 hours. Bcl-2 and Bax protein expressions for apoptosis were assessed. Cells covered 88,47% of the initial wound in the control group, 59,09% in the LPS group and 35,52% in the cytokine mixture group at 24 hours. While all three doses of resveratrol increased the wound healing in cytokine groups, in LPS groups resveratrol increased the wound healing at doses of 0,5 and 1µM. Bcl-2 and Bax protein expressions did not show any statistically significant differences between all groups. These findings suggest that resveratrol may be a candidate agent for prevention from necrotizing enterocolitis.
