In-vitro Investigation of the Effect of Corticosteroids on Candida albicans Biofilm Formation

Project no. 264 · Status: Kapanmış · 2017

Summary

Today, Candida albicans is among the opportunistic important human pathogens that usually cause superficial and mucosal infections. Most of biofilm forms are very resistant to antifungal. Therefore, biofilm production should be prevented with drug treatment. In this study, it was aimed to measure the biofilm formation of Candida albicans isolates and to analyse the effect of corticosteroids on biofilm-forming isolates. In our study, it was used the isolate of 32 Candida albicans (3 fluconazole resistant, 29 fluconazole sensitive) that isolated from blood cultures from mycology laboratory at Ege University Medical Faculty Medical Microbiology Department. First, Candida albicans isolates biofilm formation capacities were researched by using microplate method. In the next step, the effect of dexamethasone and methylprednisolone corticosteroids on biofilm production at 1000, 100, 10, 1 and 0.1 µM concentrations was analysed. As a result of our study, it was determined that 13 (41%) of 32 Candida albicans isolates produced medium level biofilm, 18 (56%) of 32 Candida albicans isolates produced low levels of biofilm and 1 (3%) of 32 Candida albicans isolates did not produce biofilm. When the effect of dexamethasone on biofilm production from corticosteroids was evaluated, while there was an increase of 3.2% to 6.4% of isolates due to the study constration and a decrease of 9.7% to 29% of isolates due to biofilm production was seen. As for 6?-methylprednisolone was determined to reduce biophilic production in 35.5 - 48.4% of isolates in different constructions. The increase in biofilm production due to 6?-methylprednisolone was realized at a concentration of only 100 µm and 3.2% rate. According to the results of our study, the effect of 6?-methylprednisolone compared to dexamethasone was determined to decrease the production of Candida albicans biofilm.
