Development of a New Glycosylation System for Some Rare Sugars

Project no. 21650 · Status: Kapanmış (Yayın bekleniyor) · 2020

Summary

The O-glycosidic bond is found in the structure of many natural compounds and drug molecules. Therefore, its efficient synthesis is a very important research area today. With this project, a new stereoselective glycosylation method has been developed to synthesize D-gulo- and D-allo- structured furanosidic glycosides.

There are many glycosylation methods in the literatüre. These methods have their own advantages and disadvantages. Some of the glycosyl donors used in these methods are compounds (eg Acetobromo-D-glucose) produced by many global chemical companies and have significant turnover. However, there is no systematic method in the literature to synthesize D-gulo- and D-allofuranosidic glycosides. Therefore, these sugars cannot be used in molecular modeling-based drug designs and drug synthesis. With the presented project, it is expected that an important step has been taken towards eliminating this deficiency.

The glycosyl donors synthesized by the project provided an important convenience for the glycosylation of some monosaccharides that are rare in nature. On the other hand, thanks to the orientation of the lattice structures of the developed method, a method with high stereoselectivity was obtained.

For this purpose, tricyclic-1,2,3-orthoester glycosyl donors of related rare sugars were synthesized. Then, the reaction conditions in which the donors were most effective in terms of reaction efficiency and stereoselectivity were found in the glycosylation reactions.
