Summary
M1 polarized and M2 polarized including two phenotypes in the cancer microenvironment, have opposite effects and are important in the progression of cancer. In the thesis study, it was aimed to polarize the macrophages in the cancer microenvironment into different phenotypes and to investigate the effects of these phenotypes on cancer cells. For this purpose, MDA-MB-231 breast cancer cells and THP-1 origin macrophage cells are co-cultured in the transwell system to determine macrophage polarization in the cancer microenvironment. The apoptotoic effect and macrophage polarization of both non-induced, LPS-induced and IL-4-induced THP-1 origin macrophages on MDA-MB-231 cells were investigated in the presence of QS-21, alum and oleuropein. Determination of the contribution of macrophage polarization of cancer microenvironment in the treatment of breast cancer, determination of the role of QS-21, alum adjuvant and oleuropein in cancer treatment and As a result of its effect on macrophages, it is aimed to contribute to the development of new treatment methods. The data obtained in the thesis study determined that CD11b expression was 99% in all study groups. The expression levels of M1 polarized macrophage marker CD86 and CD80 increase in LPS-induced groups. CD163 expression levels, which are M2 polarized macrophage markers, were generally observed to be increasing in all groups. In LPS-induced groups, there was an increase in CD86 and CD80 gene expression correlated with flow cytometry results. IL-6, TNF-?, IL-1?, IL-1ß and IL-10 cytokine levels were found to be increased in LPS-induced groups. Oleuropein exhibited anti-inflammatory effect while alum adjuvant exhibited pro-inflammatory effect. The highest apoptotic effect on MDA-MB 231 cells is determined in the group in which the substances are used together. In addition, oleuropein has been found to have a pre-apoptotic effect on MDA-MB-231 cells. As a result, QS-2, anti-inflammatory effects oleuropein and alum has an effect on macrophage polarization also, oleuropein has apoptotic effect on cancer cells and that this molecule can be effective promises in the treatment of cancer.
