Summary

The avian coronavirus infectious bronchitis virus (AvCoV-IBV) is a respiratory disease. It causes economic losses in the poultry industry and deaths worldwide. AvCoV-IBV belongs to family of viruses same as the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), and its genetic structures are highly similar. This thesis, aimed to create an in vitro antiviral model system for cytokine storm and respiratory problems caused by SARS-CoV-2 virus. Due to the fact that AvCoV-IBV is not zoonotic and provide an easier and safer working environment, experiments have been carried out with AvCoV-IBV. In the thesis co-culture model system established by AvCoV-IBV-infected bronchoalveolar carcinoma cells (CRL-5807), mesenchymal stem cell (MSC) and THP-1 cells differentiated from monocytes. The antiviral effect of Scoposide A., Scoposide B., Aristatoside B., Balansoide B., Elmalienoside A. saponin molecule combinations determined by using the tissue culture infective dose 50 (TCID 50 ) method. Supernatants from each group collected, and IL-6, TNF-? and IL-1ß cytokine were analyzed by ELISA to determine the anti-inflammatory effect. In the results of the study, a decrease in the cytokine amounts of MSC-saponin combinations calculated. When the amount of IL-6 cytokine calculated, it was determined that the combinations with Aristatoside B. and Balansoide B. saponin molecules decreased (p>0.05). The amount of TNF-? cytokine, significantly decreased in all Cephalaria spp. saponin combinations with QS-21 (p<0.0001). When the IL-1ß amounts calculated, a decreament observed only in the saponin groups containing Scoposide A. and QS-21 molecules. HA and TCID 50 results determined to measure viral titers in the antiviral model system. As a result of the experiments, it was determined that Aristatoside B., Scoposide A., Scoposide B., Elmalienoside A., Balansoide B. saponins and QS-21 molecule-MSC combinations decreased viral titer. Therefore saponin molecules can be used as promising antiviral molecules.
