In Vitro Evaluation of the Effect of Hyaluronic Acid Loaded Chitosan Nanoparticles on Bisphosphonate Treated Gingival Fibroblast Epithelial and Osteoblast Cells

Project no. 21063 · Status: Kapanmış (Yayın bekleniyor) · 2019

Summary

Within the scope of the project, it was aimed to determine the effects of hyaluronic acid-loaded chitosan nanoparticles on human gingival fibroblast, human epithelial cells and human osteoblast cells in order to examine the efficacy of bisphosphonate-induced osteonecrosis of the jaw. In this study, zoledronic acid, which is one of the nitrogen-containing bisphosphonate group drugs, hyaluronic acid, which is actively used in wound healing and accelerates the healing process by ensuring the humidity of the environment, and chitosan with antibacterial properties were used. In order to prevent the rapid dissolution of hyaluronic acid in water and to stabilize the delicate structure of chitosan, the synthesis conditions were optimized and the zeta potential of chitosan-hyaluronic acid nanoparticles was found to be +32.3 mV, and a loading efficiency of 61.625% was obtained. In order to examine the in vitro effects of the formed nanocarrier system and zoledronic acid on HGF-1, SaOS-2, HaCAT cells, cytotoxicity, apoptosis and wound model studies were carried out.
