Project no. 28474 · Status: Kapanmış (Yayın bekleniyor) · 2023

Summary

The aim of this study was to evaluate the cytotoxicity, potential for mineralization, microhardness and pH of an alkasite and a self-adhesive hybrid composite and high viscosity glass ionomer cement. Three-dimensional cultures of odontoblast-like cells and dentin were used in order to evaluate transdentinal cytotoxicity of the restorative materials within the perfusion system. Restoration materials were stored in culture medium 24 hours for cell viability MTT test. After that human dental pulp cells were incubated with this culture medium. Alkaline phosphatase test was performed after 3rd, 7th and 14th day incubation periods of restoration materials with human dental pulp cells. Similarly, Alizarin red staining was performed after 14th and 21st day. Before the application of both test methods, the indirect contact of the restoration materials with cells were carried out using the Transwell technique was used to provide indirect contact between the materials and cells. All tests were performed using spectrophotometric analysis according to their protocols. Following the culture medium contact, vickers microhardness of the restoratives were determined at initial, 1st, 3rd, 7th, 14th and 21st day. The pH change of the culture medium was observed at 1st ,3rd and 7th. All materials presented grater than %70 in transdentinal cytotoxicity findings for cell viability. Alkasite and high viscosity glass ionomer cement were not different from the negative control in the dentin barrier test (p>0.01). Cell viability MTT results indicated a significant difference between the alkasite and the high viscosity glass ionomer (p<0.01). The high viscosity glass ionomer presented the highest alkaline phosphatase activity at 3rd and 7th day (p<0.01) while it was similar to that of self-adhesive hybrid composite at 14th day (p>0.01). For alizarin red staining, high viscosity glass ionomer had higher activity than the control group at 14th and 21st days (p<0.01) while at 21st day, it's activity was not significant from the self-adhesive hybrid composite (p>0.01). The pH values for all materials were above 6.9. The difference among the microhardness measurements of the restoratives was significant (p<0.01). Dentin barrier test and cell viability MTT cytotoxicity test results do not seem well-matched with each other. Self adhesive hybrid composite had moderate cytotoxicity in the dentin barrier test. The crystallisation potential of the high viscosity glass ionomer appears to be higher than the others. Microhardness measurements did not point out the material detoriation.
