Summary

ABSTRACT:

PURPOSE: The aim of this study is to enhance the efficiency of amniotic membrane extract (AME) preparation and stabilization during the lyophilization process. The study seeks to compare the cost-efficiency of different methods used in the preparation of AME.

MATERIALS AND METHODS: Ten amniotic membranes were collected during cesarean sections, washed, cut into small pieces, and collected in a pool. The samples were first divided into two main groups based on the extraction buffer: phosphate-buffered saline (PBS) or low-salt solution. Subsequently, each group was further subdivided into two subgroups according to the presence or absence of a protease inhibitor. Following homogenization, all samples further divided into two groups based on whether trehalose was added prior to lyophilization or not. All eight groups were analyzed for total protein and growth factor concentrations to compare the efficiency of each preparation condition.

RESULTS: When growth factor concentrations were individually examined in AME samples prepared under different conditions, the effects of trehalose and protease inhibitor use varied for each growth factor. Overall, considering total protein and growth factor levels, as well as the cost-efficiency, it appears most appropriate to use PBS as the extraction buffer, to avoid protease inhibitors during extraction, and trehalose during lyophilization.

CONCLUSION: Depending on the intended clinical application, the concentrations of growth factors within AME can be adjusted to the desired levels by modifying the extraction conditions. Evaluating extraction efficiency individually for each growth factor will serve as a guide for developing personalized treatment strategies tailored to specific clinical needs.

Keywords: Amniotic membrane, amniotic membrane extract, efficiency, stabilization, lyophilization
