Summary

Cancer incidence and mortality are increasing rapidly worldwide. Chemotherapy is a frequently used treatment method, but the toxic side effects of chemotherapy reduce quality of life and survival rates. Pemetrexed (PMX) is a chemotherapy drug used in the treatment of various types of cancer. Liposomes have unique properties for the delivery of anticancer drugs. A suitable combination of chitosan and liposomes can both integrate the advantages of the system and reduce its disadvantages.

In this thesis, liposome formulations of PMX active substance were prepared, polymeric coating was carried out using chitosan (CS). The optimum amount of drug for liposomes containing PMX was determined as 6 mg/mL and the drug adsorption efficiency was determined as 68.50%. According to the results, it was determined that the particle size of the chitosan coated liposomes was 220±5 nm, the polydispersity index was 0.22±0.03 and the zeta potential was 23.6±1.40 mV. In vitro drug release tests were carried out at pH levels of 5.5 and 7.4 with pemetrexed- loaded chitosan coated liposomes (CS-PMX-LİP) and free pemetrexed. The results obtained showed that liposomes has a controlled drug release profile.

As a result, the developed liposome system has the potential to be investigated in vitro and in vivo in further studies to increase pemetrexed's controlled release and therapeutic effect.
