Aqueous Humor Transcriptome Analysis in Eyes with Exfoliative Glaucoma
Project no. 29593 · Status: Kapanmış (Yayın bekleniyor) · 2023
Summary

Purpose: To determine the differentially expressed genes in aqueous humor (AH) of eyes with pseudoexfoliation syndrome (PXS), exfoliation glaucoma (XG) and healthy eyes and to analyze the microbiome of aqueous humor of these eyes.

Methods: Treatment naïve, newly diagnosed patients with PXS and XG in both eyes and healthy individuals with no systemic or ocular conditions/medications will be included in the study. AH collection will be performed during cataract/glaucoma surgery. DNA and RNA extraction from AH will be performed using commercial kits. mRNAs will be isolated and used in library preparation. Transcriptome sequencing will be performed using Nanopore third generation sequencing platform. Differentially expressed genes will be validated by qPCR. Extracted DNA will be subjected to bisulfite conversion. Promoter methylation status of DEGs will be investigated by qPCR. For bacterial microbiome, extracted DNA will be used to prepare 16S sequencing libraries. They will also be sequenced by using Nanopore third generation sequencing technology. Analysis of the data will be carried out by professional bioinformaticians.
