Identification of Helicobacter pylori HP1437 protein as a TIR domain protein, its recombinant production, purification and investigation of interactions with MyD88/TIRAP

Project no. 534 · Status: Kapanmış · 2015

Summary

The aim of the project was to identify and characterize HP1437 as a bacterial TIR domain protein from H.pylori. H.pylori is an important pathogen which causes several gastric related diseases, including gastric cancer. H.pylori manages to hide from immune system and thus can survive in gastric mucosa for many years. In this Project, it was proposed that H.pylori, similar to other pathogens, might have TIR domain proteins which can manipulate immune system. Bioinformatic searches revelad HP1437 as a putative TIR domain protein. HP1437 was expressed and produced as a recombinant protein, purified and its structurally stable domains were characterized. The in vitro interaction of the purified HP1437 with MyD88 was shown. The protein –protien interaction between HP1437 and mMyD88 supports that HP1437 can manipulate TLR signal transduction by binding to human TIR domain adaptors. In parallel to experimental studies, the structure of HP1437 was modelled by homology modelling and the presence of TIR domain structure was shown. In summary, all these support that H.pylori posses a TIR domain protein and this protein was, fort he first time, characterized by bioinformatics and biochemical methods.
