Significance of lncRNA’s in Diagnosis and Prognosis of Pancreatic Cancer
Project no. 18 · Status: Kapanmış · 2016
Summary
Pancreatic cancer is the fourth most common type of cancer-related mortality. Certain antigens have been identified in the serum of patients with pancreatic cancer. Various molecular studies including small noncoding RNA such as siRNA, miRNA ve lncRNA have been investigated as a new biomarker of pancreatic cancer. The aim of this study was to investigate the roles of lncRNAs in pancreatic cancer and to demonstrate the relation between lncRNA levels and clinical, demographic and labaratory findings of pancreatic cancer patients. In our study, normal pancreatic (HPDE) and pancreatic ductal adenocarcinoma (PANC-1) cell lines were used. In addition, 40 patients with pancreatic cancer and 10 healthy individuals were included in the study. In our study, the expression leves of lncRNAs in patients with pancreatic cancer were compared with the expression leves of cell series samples and whether these could be a diagnostic test and prognostic indicator. Analysis of pancreatic cancer cell line showed that ANRIL, H19, HOTAIR, MALAT1, PCA3 and PTCSC3 expression levels increased 0.58 fold, 4.60 fold, 4.19 fold, 0.66 fold, 1.25 fold and 0.38 fold, respectively. When the change of the same lncRNAs in patient plasma samples compared to healthy controls was observed, it decreased 5.85 fold, 3.91 fold, 4.22 fold, 1.70 fold, 1.81 fold and 3.93 fold, respectively. It was found that plasma lncRNA changes in patients with pancreatic cancer were affected by factors such as alcohol and cigarette smoking and diabetes. In addition, as the disease stage increases, plasma lncRNA changes increase. As a result, the expression of H19 and HOTAIR lncRNAs in PANC1 cell line was increased in vitro analyzes. However, ANRIL, H19, HOTAIR and PTCSC3 lncRNA levels are decreased in plasma samples of patients with pancreatic cancer. Since this change is more apparent in advanced disease, smoking, alcohol use and diabetes, it is not possible to use it as a diagnostic method in the early diagnosis of pancreatic cancer.
