Pilot scale production and formulation of probiotic bacteria

Project no. 21412 · Status: Kapanmış · 2019

Summary

In this project, optimum growth conditions of Lactobacillus plantarum BG24 strain isolated from boza were determined and scale-up studies were carried out. The biomass obtained from L. plantarum BG24 strain was dried by spray dryer and a probiotic food supplement prototype was obtained. The probiotic properties of lactic acid bacteria (LAB) isolated from different foods were also investigated. The probiotic potentials of the LAB isolates were calculated and the strain isolated from boza which was named as L. plantarum DGA55 were found to have the highest probiotic potential (90 %). The specific growth rate, the doubling time and the biomass productivity of L. plantarum BG24 in optimized MRS broth (pH 6.5) medium enriched with 5 g/L yeast extract was found to be 0.483 s-1, 1.43 s and 0.17 gL-1s-1, respectively. The optical density, biomass concentration, number of viable cells were found to be 9.6 OD600, 3.8 g/L and 7.6×1011 cfu/mL, respectively in the 5-liter bioreactor experiments under the optimized conditions. Scale-up studies were performed in a 10-liter stirred tank bioreactor based on power count calculation. In the pilot scale 100-liter stirred tank bioreactor (agitation rate 270 rpm) the optical density, biomass concentration, viable cell count was found as 8.2 OD600, 2.75 g/L and 1.4×1010 cfu/mL, respectively. Finally, L. plantarum BG24 and DGA55 strains were spray dried and enteric capsules were produced. It was determined that the number of probiotic bacteria in the capsule decreased from 8.37±0.03 log cfu/250mg to 6.16±0.40 log cfu/250mg during the 6-month storage period at 25°C.
