Evaluation of New Generation Platelet Concentrates in Bone Regeneration and Reconstruction

Project no. 21708 · Status: Kapanmış (Yayın bekleniyor) · 2020

Summary

Aim: The aim of this experimental study was to assess whether advanced- and injectable-platelet rich fibrin (PRF) enhance effect of beta tricalcium phosphate (ß-TCP) on repair of bone defects.

Materials and Methods: This experimental study was conducted on fourteen, 6-8 months old, 2-2.5 kg of avarage weight, New Zeland rabbits. The animals were operated in both femur to create 3 mm diameter four bone defects. These defects were divided into subgroups as follows: injectable-PRF+ß-TCP, advanced-PRF+ß-TCP, ß-TCP alone and empty defect as control group. The animals were sacrified at postoperative 4th and 8th weeks, and bone samples were collected. Histological analysis was performed using hematoxylin-eosin staining, and fibronectin/bone sialoprotein immunoreactivity were evaluated to identify the cells in healing tissue. Micro-CT was used to analyse bone volume and bone mineral density.

Results: Histological analysis at 4th and 8th weeks revealed that lamellar bone formation was only found in injectable-PRF+ß-TCP group. In the injectable-PRF+ß-TCP group, fibronectin was found at a low rate (p<0.001), while the bone sialoprotein ratio was found to be higher than the advanced-PRF+ß-TCP and ß-TCP alone groups (p<0.001). Micro-CT analysis showed higher bone volume and bone mineral density in injectable-PRF+ß-TCP group compared to advanced-PRF+ß-TCP and ß-TCP alone groups at both 4th and 8th weeks (p<0.001).

Conclusion: The combination of injectable-PRF and ß-TCP showed superior outcomes compared to A-PRF or ß-TCP alone. Therefore, it is suggessted that this PRF matrice could be useful as a supplementary material to support the bone formation.
