PREPARATION OF QUERCETIN AND CHLORHEXIDINE LOADED PLGA FORMULATIONS AND DEVELOPMENT OF PROTOTYPE PRODUCT FOR THE TREATMENT OF PERIODONTAL DISEASES

Project no. 22540 · Status: Kapanmış · 2020

Summary

Periodontal diseases are inflammatory diseases that occur as a result of interactions between microbial dental plaque settled in the gingival groove and host responses. Our project aims to develop a prototype product of the quercetin and chlorhexidine loaded PLGA system to be applied locally in the treatment of periodontitis and to examine its antimicrobial and anti-inflammatory bidirectional effectiveness in vitro and in vivo. For this purpose, chlorhexidine (CHX), which is widely used in oral care products due to its broad-spectrum antimicrobial properties, and quercetin (QUE), which is a natural product known for its antibacterial activity as well as its anti-inflammatory and wound healing accelerating properties, were used in different amounts (1.5, 5 and 15%). The CHX and QUE active substances were loaded single or binary into Poly(D,L-lactic-co-glycolic acid) nanoparticles (PLGA NP) with high efficiency, and their structural, morphological and thermal properties were characterized. In-vitro release studies of CHX/ QUE loaded nanoparticle were carried out in artificial saliva environment, and the amounts of CHX and QUE were determined by high-pressure liquid chromatography (HPLC). A partial validation study was carried out for the active substances CHX and QUE, and the results met the specified criteria. When the Dynamic Light Scattering (DLS) results of PLGA NPs loaded with the synthesized CHX/ QUE active ingredients were examined, it was seen that the dimensions varied in the range of 100-150 nm. When the stability test data performed under +4 oC conditions were evaluated, it was concluded that both active substances were stable. While the QUE active ingredient showed a release of 168 hours when loaded alone in the artificial saliva environment, it was found that when the CHX/QUE active ingredients were loaded dual, it showed an effectiveness of 20% even at the end of the 240th hour. As for the active ingredient CHX, it is observed that it is more than 70% effective even at the end of the 240th hour for both single and dual systems. In addition, an experimental periodontitis disease model was developed at the end of the 21st day by silk suture materials tied around the second molars in the upper jaw of Albino Wistar rats.

This project work was supported by the Ege University Scientific Research Project numbered TUA-2020-22540.
