FERMENTATIVE PRODUCTION, PURIFICATION AND CHARACTERIZATION OF L-ASPARAGINASE ENZYME FROM Pseudomonas sp.

Project no. 663 · Status: Kapanmış · 2017

Summary

L-asparaginase aminohydrolase (L-asparaginase EC 3.5.1.1) is a high value-added enzyme in many industries by means of providing an alternative treatment to various types of cancer, such as acute lymphoblastic leukemia, malignant lymphoma and preventing the formation of acrylamide that is a neurotoxin in foods processed at high temperatures. The enzymes obtained from the prokaryotes are used in therapeutical asparaginase production, such as E. coli and E.chrysanthemi, can cause hypersensitivity and coagulation defects in the human body. New asparaginase producing microbial strain screening studies that do not show these side effects are becoming increasingly important. Within the scope of this research project, L-asparaginase producer strain was determined among various Pseudomonas species and fermentative production of L-asparaginase with different carbon and nitrogen composition in shaking cultures. Produced enzyme was purified by using basic techniques in biochemistry. After isolation and purification steps, L-asparaginase enzyme was characterized. Moreover, the cytotoxic effects of the purified enzyme was investigated. Key words: asparaginase, fermentative production, izolation and characterization of enzyme, cytotoxic activity.
