Summary

Bacteriophage therapy is gaining importance again in the era of antibiotic resistance. Bacteriophages are viruses that infect bacteria and multiply in this way. The presence of these microorganisms, which are widely found in all ecosystems where bacteria are found, in the human body has been known for many years. Within the scope of this thesis; It is aimed to evaluate the isolation, characterization and the potential use of lytic bacteriophage from patient respiratory samples against A. baumannii, which is classified in the critical priority group by the World Health Organization in terms of the development of new antimicrobial treatments. Fifty deep tracheal aspirate, transtracheal aspirate, bronchoalveolar lavage fluid and bronchoscopic aspiration fluid samples sent for routine microbiological examination to the Ege University Faculty of Medicine Medical Microbiology Bacteriology Laboratory between April 17 and July 9, 2023 and from which A. baumannii was isolated were considered as potential bacteriophage sources and included in the study. The presence of bacteriophages in these sources; was investigated by spot test method on A.baumannii isolates isolated from the same samples. Lytic bacteriophage was isolated against an A.baumannii isolate. When the bacteriophage was examined with an electron microscope, it was seen that it was a tailed phage belonging to the Caudoviricetes class. The host range was investigated on A.baumannii isolates isolated from patient samples included in the study and the A.baumannii ATCC BAA-1605 strain. It was seen that the bacteriophage, which was not effective against these strains, had a narrow host range. In taxonomic analysis, it was observed that 79% of the bacteriophage genome was 98.80% similar to the previously identified Acinetobacter phage WCHABP1 in the Obolenskvirus genus. It is thought that the bacteriophage can be classified as a new member of the Obolenskvirus genus. In vivo testing of bacteriophage containing gene regions associated with lysogeny is necessary to evaluate the safety of treatment.
