Development of a Controlled Drug Delivery System for the Treatment of Periodontitis
Project no. 23471 · Status: Kapanmış (Yayın bekleniyor) · 2022
Summary

Within the scope of this project, a system providing controlled and combined drug release for the treatment of periodontal diseases was developed. Poly(lactic acid) (PLA)-based nanogels functionalized with acrylate and glucosamine and loaded with doxycycline and simvastatin, possessing antibiotic and anti-inflammatory properties, were successfully prepared. PLA polymers functionalized with glucosamine to provide bioadhesive properties and with acrylate groups to confer antimicrobial activity were synthesized and combined at a 50% ratio to obtain nanogel formulations using the emulsion–solvent evaporation method. The prepared nanogels were evaluated under in vitro conditions in artificial saliva for their 72-hour release kinetics, stability, sterilization compatibility, and bioadhesion properties. Their antimicrobial activities were investigated against P. gingivalis, S. aureus, E. coli, P. aeruginosa, E. faecalis, and A. actinomycetemcomitans, and their minimum biofilm inhibitory concentrations and effects on biofilm formation were determined. Based on the project outcomes, in vivo testing of the developed product at the Forsyth Institute (USA) was planned, and periodontitis animal models were established for this purpose.
