Kudret narının (Momordica charantia L.) Ratlarda Trinitrobenzensülfanik asit (TNBS) ile oluşturulmuş deneysel kolit modelinde etkinliğinin araştırılması

Project no. 80 · Status: Kapanmış · 2017

Summary

Aim:Momordica charantia L. known as bitter melon (BM) belongs to Cucurbitaceae. The aim of this study is to investigate anti-inflammatory effect of BM on TNBS-induced colitis in rats. It’s also aimed to determine the bioaccessibility of total carotenoid and ß-carotene of BM. Methods:The bioaccessibility was performed with in-vitro digestion. Rats were divided into 4 groups; BM+TNBS, BM, TNBS, control. BM was given 300 mg/kg/day for 6 weeks orally. Colitis was induced by 2%TNBS in 50%ethanol(w/v) intrarectally after 6 weeks. After 7 days, animals were sacrificed; macroscopic-microscopic examinations were performed. Myeloperoxidase, IL-17, TNF-alpha, IL-10 were measured byELISA. Results:The bioaccessibilities of carotenoid and ß-carotene were 2.12% and 0.82%, respectively. BM ameliorated macroscopic-microscopic colitis damages, (p=0.015), (p=0.026). Anti-inflammatory cytokine IL-10 significantly increased in serum in BM+TNBS and BM groups (p=0.000). Conclusion:BM has a protective anti-inflammatory effect on colitis. This effect might be related to both carotenoids and the other bioactive compounds of BM. Keywords: Momordica charantia, anti-inflammatory, TNBS-induced colitis, digestion, carotenoid
