OPTIMIZATION OF CELLULASE PRODUCTION BY SOLID STATE FERMENTATION USING LOCAL ISOLATE OF ACTINOBACTERIA

Project no. 22356 · Status: Kapanmış · 2020

Summary

In this study, it is aimed to optimize cellulase production from local Acrinobacteria isolate by solid state fermentation method and partial characterization of the enzyme. Cellulase is used extensively in various industrial processes and is produced at high capacities worldwide. Its wide usage potential necessitates the production of cellulases with different qualities. In this thesis, M127-1, M127-2B, 1M1, M45-1 and M6c-1 isolates were used among actinomycete strains, which were previously screened and determined to be good cellulase producers. These strains were inoculated into nutrient medium containing wheat bran and malt grass, and it was determined that the strain that produced the best cellulase was M127-1. In the next step, M127-1 was observed to grow on media containing different combinations of sawdust, malt grass and wheat bran. The best growth and cellulase activity were obtained with a medium containing wheat bran and malt grass. Then, using this nutrient medium, the parameters of humidity, inoculum amount and incubation time were optimized with statistical Experimental Design approach. As a result of the optimization, the best cellulase production conditions were determined as 79.93% humidity, 8.58 days incubation time and 8.38 (v/w) inoculum amount. The incubation time had a higher effect than the other two parameters and the enzyme activity was increased 1.8 times under optimized conditions. The optimum pH of the enzyme is 4, the optimum temperature is 60°C and the best pH stability is pH 6.In the shelf life examination of the lyophilized culture liquid, cellulase remained activity at room temperature by 58% at the end of the 4th week. According to 16S rDNA analysis of the M127-1 isolate, it was determined that it is a member of Streptomyces. In this thesis, production optimization of cellulase, which is active at acidic conditions and high temperature, by solid state fermentation using cost-effective substrates with Streptomyces sp. M127-1 isolate was carried out.
