THE EFFECTS OF BIBR1532 AND RESVERATROL COMBINATION ON COLORECTAL CANCER TYPES AND WNT SIGNAL PATHWAYS

Project no. 23172 · Status: Kapanmış · 2021

Summary

Colorectal cancer (CRC) is the second most commonly seen cancer type in humans. This situation proves that it is important and crucial to study the molecular mechanisms of CRC. WNT is also recognised as a distinct molecular pathway on most cancer types. Therefore it can be said that, the hyperactivisation of the WNT signal pathway is a supportive force for CRC. Resveratrol that works as a natural polyphenol compound, has anti-oxidant and anticancer effects. BIBR1532, on the other hand is known to be a telomerase inhibitor. This study aims to research the effects of the combination of BIBR1532 and resveratrol on HT29 cells which are the cells of CRC, WNT signal pathways. Aiming the evaluation of the effects of these two agents on colorectal cancer cells, WST-1 test and IC⁵⁰ doses were determined. The combination indexes were figured by isobologram analysis and the mentioned doses were applied in the cells to actualise the total RNA isolations in our prior studies. This study also consists of cDNA in qRT-PCR analysis and WNT signal pathways were determined by 2-∆∆CT method. According to the expression results, BIRB1532 and resveratrol combination were proved to up-regulate many genes functioning as tumor suppressor in WNT signal pathway as well as down-regulating most of the genes that function as oncogene or proto-oncogene by quantitative results.
