DETERMINING THE POTENTIAL TOXIC EFFECTS OF SOME PARABEN DERIVATIVES BY MEASURING THE EMBRYOTOXICITY

Project no. 371 · Status: Kapanmış · 2018

Summary

Parabens started to be used for the first time in the 1920s as a preservative in pharmaceuticals and increased production and use of preservatives in many food and industrial materials, especially in cosmetics. Despite the rapid degradation in aquatic environment, paraben varieties which are more permanent chlorine derivatives which are degradation products can accumulate in sediments and living organisms. Although studies have been conducted in various human cell cultures and experimental animals, studies on their effects on aquatic organisms are scarce. In the current study the effects of parabens derivatives on sea urchin using Arbacia lixula species was investigated. The embryotoxic, spermiotoxic and genotoxic effects of Butylparaben, ethylparaben, propylparaben and benzylparaben, which are 4 paraben derivatives, have been demonstrated. Millions of eggs increase the reliability of the experiments statistically. Furthermore, sea urchin embryos provide the opportunity to work with a living metazoon. These trials; cheap, short-term, reliable and sensitive are important features. In embryotoxicity experiments, deterioration occurred in sea urchins that passed to larval stage after fertilization and were exposed to paraben varieties, while in experimental groups obtained by exposure of sperm; fertilization success and larval deterioration after fertilization were taken into consideration. In genotoxicity experiments, embryos exposed to paraben derivatives after fertilization were fixed after 5-6 hours and chromosome damage was determined. When the embryotoxicity results were compared according to the data obtained with this project study, it was found that Benzylparaben was 0.446 mg / L and Ethylparaben was 17.558 mg / L. According to the data obtained from spermiotoxicity experiments; The highest toxic effect is seen in Benzylparaben with 4.66 mg / L. When the genotoxic effect was taken into consideration, the biggest effect was found in Butylparaben. When all the data are evaluated together, it can be asserted that paraben derivatives exert pressure on the marine environment.
