Synthesis and Biological Activity Studies of Some Phthalimide-sulfonamide Derivatives as Potential Alpha-Glucosidase Inhibitors
Project no. 23647 · Status: Kapanmış · 2022
Summary

Diabetes is a chronic metabolic disorder characterized by high blood sugar level, which occurs as a result of insufficient production of the insulin hormone or inability to use the produced insulin effectively. Diabetes is seen as the most important health problem for patients and society with the highest mortality, morbidity and cost worldwide. For such reasons, it has been demonstrated that diabetes treatment should be carried out in a controlled manner.

In the treatment of diabetes, it is aimed to control blood glucose with different mechanisms of action. For this purpose, α-glucosidase inhibitors are used to prevent postprandial hyperglycemia. When α-glucosidase inhibitors are used, glucose absorption is reduced and postprandial hyperglycemia is prevented without causing hypoglycemia.

There are three drugs used as inhibitors of α-glucosidase, these are acarbose, miglitol and voglibose. Although these drugs are used in treatment, their high cost synthesis, moderate efficacy and gastrointestinal side effects make their long-term use difficult. For all these reasons, compounds with different core structures have been studied in order to develop more reliable and more effective new α-glucosidase inhibitors, including phthalimide and sulfonamide compounds. Based on this information, three new phthalimide-sulfonamide derivative compounds that are potential α-glucosidase inhibitors were designed and synthesized. The structures of the synthesized compounds were confirmed by IR and 1H NMR spectroscopy methods. The α-glucosidase inhibitory activities of the compounds were tested in vitro using the spectrophotometric method. According to the biological activity results, the compounds showed higher inhibitory activity than the reference compound acarbose at varying rates.

Keywords: Phthalimide; Sulfonamide; α-Glucosidase Inhibitors; Synthesis
