DEVELOPMENT OF A METHOD FOR ANALYSIS OF GANGLIOSIDES WITH CAP-LC-ESI-MS/MS AND APPLICATION IN BIOLOGICAL SAMPLES
Project no. 448 · Status: Kapanmış · 2017
Summary
Gangliosides are glycosphingolipids bearing one or more sialic acids (Sia) and complex sugars. Ganglioside molecules are found in various biological fluids (blood and milk) and tissues (nerve, muscle, brain). The Liquid Chromatography-Mass Spectrometer (LC-MS) system has been preferred for the determination of gangliosides in biological samples due to its highly sensitive and selective analysis capability. First, the method for gangliosides using LC-MS / MS system was developed with standard material. The extraction method was then optimized using milk. For this purpose, liquid-liquid extraction with chloroform / water / methanol mixture and then solid phase extraction (SPE) were used. The developed method was applied to Bombyx mori, Galleria mellonella and Tenebrio molitor tissues used as insect model organisms. Gangliosides were detected as a result of milk extraction, but no gangliosides were detected in insects. Based on these results, it has been found that these molecules, which are controversial for insects, are difficult to detect by conventional methods. It is thought that more advanced methods should be performed with the support of genetic tools for further research.
